Gillespie was born in Armagh to a family of Scottish descent; and trained in Dublin and later Edinburgh. He joined the Royal Navy as a surgeon in 1781 and travelled extensively; and in 1805 he was officially attached to HMS Victory but was ashore suffering from gout on the day of the battle of Trafalgar. He was also inspector of Naval Hospitals in Malta; Sicily; Gibraltar and Naples. He retired from the navy in 1819; and settled in Paris. He was the author of two books on tropical diseases.
